Equipment

The Ad-Light Microscopy Core Facility has a microscope with structured illumination (SIM), which is ideal for super-resolution fluorescence microscopy, and a Zeiss Axio Observer, which is equipped with a camera and can be used for wide-field fluorescence microscopy. In addition, two confocal laser scanning microscopes (LSM) with heating unit and CO2 module are available. The Zeiss LSM880 is additionally equipped with an AiryScan detector. Users of the Ad-Light have access to a workstation that enables AI-based, automated, quantitative image analysis of large microscopy images and image data sets.

The Zeiss LSM 880 with Airyscan detector is an advanced, versatile confocal microscope. It is suitable for microscopy of both vital cells and fixed samples. The microscope is suitable for the detection of all common fluorochromes. It has a heating unit and a CO2-module and is therefore optimally equipped for live cell experiments. The integrated Airyscan detector enables high-resolution fluorescence microscopy, increasing the conventional optical resolution by a factor of 1.7. The innovative Airyscan detector, which can be operated in “Fast Airyscan” mode, makes it possible to record rapid processes in vital cells with high sensitivity and resolution. It is also suitable for specialized examination techniques such as FRAP, FRET and FCS.

The Zeiss LSM700 is a confocal laser scanning microscope which is well suited for imaging vital and fixed cells or tissue sections. The device is equipped with 4 lasers and allows the detection of all common fluorochromes. The ease of use and high sensitivity allow the creation of high-quality confocal microscopy images.

Equipment for live cell microscopy 

The LSM880 AiryScan is equipped with a CO2 module and an incubation unit.

The Zeiss Elyra PS.1 is a fluorescence microscope that can utilize various techniques to produce super-resolution images. These include structured illumination (SIM) and single molecule localization (SML) techniques such as photoactivation localization microscopy (PALM)/ Stochastic Optical Reconstruction Microscopy (STORM). This microscope is particularly suitable for the examination of fixed cells. In addition, fluorescence signals can be visualized in the immediate vicinity of cell membranes using TIRF microscopy.

The Zeiss Axio Observer Z1 is a powerful and versatile wide-field fluorescence microscope. It is very well suited for the analysis of vital and fixed cells and tissue sections. Both high-contrast transmitted light (phase contrast and differential interference contrast (DIC)) and fluorescence images can be captured and combined. The Zeiss Axio Observer Z1 enables microscopy of vital cells at constant atmosphere (temperature and gassing) to observe dynamic processes such as proliferation, cell death, motility or cell interactions in real time.

The AI workstation offers users various segmentation options for the automated quantitative analysis of large microscopy images and image data sets. This allows objects to be objectively detected and reproducibly quantified using histogram segmentation, machine learning or deep learning. The Ad-Light provides access to the ZEISS arivis Cloud, in which AI models based on neural networks can be trained for image analysis. The user-friendly interface allows both beginners and experts to use AI for image analysis.
